บทคัดย่อ/คำอธิบาย :

An optimized multiplex-polymerase chain reaction (m-PeR) assay has been developed using novel genes obtained by bioinformatic analysis. A pan-Salmonella gene (invA) and a pan-bacteria gene (l6S rRNA) served as positive controls. The mpeR assay can simultaneously detect all 6 Salmonella serotypes in a single test for S. Typhi, S. Paratyphi A, S. Paratyphi Band S. Paratyphi e, s. Enteritidis, and S. Typhimurium. The m-PeR assay showed 100% analytical specificity in identifying a panel of 33 bacterial strains. The assay's analytical sensitivity (limit of detection) was 10 2-10 3 CFU/ml or > 10 pg/ul of genomic DNA. The diagnostic sensitivity was 100% and the diagnostic specificity was 100% when tested using a panel of 190 clinical isolates consisting of S. Typhi (n=82), s. Paratyphi A (n=28), s. Paratyphi B (n=lO), S. Paratyphi C (n=1), S. Enteritidis (n=27), s. Typhimurium (n=15), other known Salmonella (n= 18), and non-Salmonella (n=9) enteric bacterial strains.
