บทคัดย่อ/คำอธิบาย :

Satu saluran baru untuk mengatasi masalah kekurangan bahan Eurycoma longifolia untuk kajian anti-kanser boleh dicapai dengan kaedah kultur ampaian se!. Untuk mendapatkan kultur ampaian sel dengan biojisim sel dan aktiviti sitotosik yang tinggi, pemilihan sel turunan elit, manipulasi keadaan kultur dengan pengoptimuman nutrien medium telah dijalankan. Keputusan kajian menunjukan bahawa sel turunan Eu6 dengan ketumpatan inokulum permulaan sebanyak 0.5 9 memerlukan 19 hari untuk mencapai biojisim sel dengan aktiviti sitotosik yang tinggi. Keadaan optimum kultur abiotik, penambahan bahan elisitor dan pengubahsuaian nutrien medium dapat merangsang penghasilan biojisim sel yang lebih tinggi sebanyak 33.9 % dan aktiviti sitotosik yang lebih tinggi sebanyak 81.7 % berbanding dengan pengkulturan sel biasa. Kaedah ekstrakan baru telah direkakan untuk mendapat ekstrak mentah dengan aktiviti sitotosik tinngi daripada sel-sel kering melalui ultrasonikasi dalam campuran methanol dan kloroform. Ekstrak mentah itu yang kemudiannya dibahagikan antara air dan etil asetat untuk mendapatkan pecahan etil asetat untuk diuji terhadap beberapa sel turunan kanser manusia in vitro. Keputusan menunjukkan ekstrak tersebut mempunyai aktiviti sitotosik yang tinggi sebanyak 11.6 IJg/mL terhadap sel turunan kanser HCT116. Hasil daripada kromatografi lapisan nipis menunjukkan sebatian dalam ekstrak mentah sel kering adalah sama dengan beberapa sebatian daripada ekstrak mentah dari akar, batang dan daun tumbuhan. A new avenue to overcome problem of insufficient Eurycoma /ongifolia materials for anticancer study could be achieved via cell suspension culture method. To obtain cell cultures with high cell biomass and high cytotoxic activity, selection of elite cell line, manipulation of culture conditions with optimization of medium nutrients were carried out. Results indicated that cell line Eu6 with an initial inoculum density of 0.5 g required 19 days of growth for ideal cell biomass production with high cytotoxic activity. The optimal abiotic culture conditions, supplement of elicitors and modification of medium nutrients were able to induce production of higher cell biomass of 33.9 % and higher cytotoxic activity of 81.7 % compared to normal suspension cell culture. New extraction method was established to obtain crude extracts from the dried cells via ultrasonication in a mixture of methanol and chloroform. The crude extract was then partitioned between water and ethyl acetate to obtain the ethyl acetate fraction that was tested against several human cancer cells in vitro. Results obtained showed the cell crude extract had high cytotoxic activity of 11.6 /Jg/mL against HCT116 cancer cell line. Results from the thin layer hromatography showed that the compounds within the crude extracts of the dried cells were similar to several compounds of the crude extracts derived from root, stem and leaf of the plant.
